Spectroscopy for Organic Structure Determination — the complete deep dive

Four spectroscopic methods, used together, take an unknown to a structure. Mass spectrometry gives the molecular mass, formula, and skeleton clues from fragmentation. IR is a functional-group detector. UV-Vis reports conjugation. NMR (¹H and ¹³C) is the connectivity engine.

0. The four-technique workflow

Each method answers a different question; the power is in combining them, always anchored to the molecular formula's degree of unsaturation (DBE) (see the Characterization guide).

TechniquePhysics probedWhat it tells you
MSmass/charge of ionsmolecular mass, formula (HRMS), heteroatoms (isotopes), skeleton (fragments)
IRbond-vibration energiesfunctional groups present/absent
UV-Visπ→π*, n→π* electronic transitionsextent of conjugation, chromophores
¹H / ¹³C NMRnuclear spin in a fieldnumber & type of H/C environments, connectivity, stereochem
The three questions to drive any elucidation: (1) What is the formula & DBE (MS + combustion)? (2) Which functional groups (IR, plus UV-Vis for conjugation)? (3) How are the pieces connected (NMR shift + integration + coupling; ¹³C for the carbon count)? Cross-check every proposed structure against the DBE and against all spectra simultaneously — a structure must fit everything.
